Antibody-Oligonucleotide Bioconjugations
The need to develop an efficient and precise detection technique for multiple, low-abundance
protein has played an important role in improving a host of biologic applications
e.g. biomarker discovery, clinical diagnostics, protein interaction network mapping
and many others. While multiplex protein detection and quantification have been
constrained by factors including assay specificity, sensitivity, and throughput.
Bio-Synthesis offers custom antibody-oligonucleotide conjugates which permit highly
sensitive immunoassay by immobilizing analytes for each conjugate onto a solid support
with cognate capture antibodies. Binding of the antibody-oligonucleotide conjugate
to the immobilized analytes allowed for amplification of the attached DNA. Products
of amplification were visualized using gel electrophoresis, thus denoting the presence
of bound analytes. Oligonucleotide-antibody conjugates have been used as suitable
tools for high-sensitive protein detection.

Immuno-PCR
An antigen detection system in which the polymerase chain reaction (PCR) is used
to amplify a segment of marker DNA that has been attached specifically to antigen-antibody
complexes. Because of the enormous amplification capability and specificity of PCR,
immuno-PCR allows considerable enhancement in detection sensitivity of a specific
antigen over conventional antigen detection systems, such as enzyme-linked immunosorbent
assays and radioimmunoassay. A variety of superior characteristics and the versatility
of immuno-PCR, including high but controllable sensitivity, simplicity, and easy
contamination control, offer great promise for its application in various aspects
of biological and medical sciences, as well as clinical diagnostics and forensic
medicine.
Multiplexed Assay Development
A planar array or bead-based arrays which allow the ability to simultaneously measure
in a quantitative manner multiple analytes of interest across multiple sample using
universal capture technology, the technology take the advantage of the specific
binding between two complimentary strand of DNA, one strand place at specific location
on the microtiter plate. Complementary oligos are conjugated to the user selected
antibody. The oligo-antibody conjugates are pooled and hybridized to the capture
oligos in the well to create an antibody array.

Chemistries
Antibody is thiolated with 2-iminothiolane and then reacted with a maleimide-activated
oligo.
Note:
In this case sulfhydryl functional groups will be introduced to an antibody through
Lys residues using a reliable and reproducible procedure. However, if some of the
modified or conjugated Lys residues are located on the antigen binding sites, this
method may produce partially active or inactive antibody-enzyme conjugates that
may not bind to the antigen. In such a case an alternative conjugation method may
be used.
Quality Control Criteria
The custom prepared product is quality controlled by Bio-Synthesis according to
our in-house standards. These are tailored for each individual assignment. The custom
conjugate is approved only when both sets of quality control criteria have been
met.
Custom Formulation
Our conjugation is based on a mixed short length polymer cross linked covalent bonded
technique. We require ligands with either a primary amine or are biotinylated.
Prices:
Due to the nature of each project, a written quotation will be made stating cost
and time required after receipt of the antibody sample.
Standard price covers labor and materials including the cost of maleimide-activated
oligo preparation, antibody thiolation, conjugation procedure, conjugate purification
and analysis.
Delivery
Conjugations take 1-2 weeks. All shipments are sent Fed Ex Standard Overnight delivery.

Sample Submission Guideline
As a rule, Bio-Synthesis will only accept pure substances as starting material,
either lyophilized or dissolved in a medium of known chemical composition. Interfering
substances must not be present. Non-commercial antibodies supplied by the customer
should be affinity purified. Please provide gel electrophoresis data along with
your antibody. Commercially available antibodies can be supplied by customers or
ordered through Bio-Synthesis. The cost of acquiring antibodies will be added to
the invoice plus an administration charge (15% of total cost). We can assist in
antibody modification or fragmentation prior to any immobilization reaction.
