Bio-Synthesis's Blood Genomic DNA Isolation Micro Kit is designed for the rapid preparation of genomic DNA from up to 1500 µL of whole blood, buffy coat, plasma and serum.

Purification is based on spin column chromatography using Bio-Synthesis's proprietary resin as the separation matrix. Briefly, the sample is lysed and the released genomic DNA is bound to Bio-Synthesis's column in the presence of Binding Solution (BIND). Under these conditions only the genomic DNA is bound to Bio-Synthesis's resin while most of the contaminating RNA and cellular proteinaceous components are removed in the flowthrough. The bound DNA is then washed to remove any remaining impurities (WASH). Lastly, the purified genomic DNA is eluted into 100 - 200 µL of the provided Elution Buffer or water (ELUTE). The Blood Genomic DNA Isolation Micro Kit allows for the isolation of genomic DNA from the blood of various species, including humans. Typical yields of genomic DNA will vary depending on the cell density of the blood sample. Preparation time for a single sample is less than 30 minutes, and each kit contains sufficient materials for 50 preparations. The purified genomic DNA is fully digestible with all restriction enzymes tested, and is completely compatible with downstream applications including PCR and Southern Blot analysis.

Genomic DNA was isolated from two different 150 µL whole blood samples using Bio-Synthesis's Blood Genomic DNA Isolation Micro Kit and a leading competitor's kit. Following isolation, 10 µL from 200 µL elution was loaded on 1% TAE agarose gel. Bio-Synthesis's Blood Genomic DNA Isolation Micro Kit demonstrated a better DNA yield than the leading competitor's kit. Lane M: Bio-Synthesis's HighRanger 1kb DNA Ladder.
Figure 2. Purified DNA Can be Amplified in a Real-time PCR (SYBR Green) Reaction. Genomic DNA was isolated from 150 µL of whole rat blood ( Δ ) and whole rabbit blood (O) samples using Bio-Synthesis's Blood Genomic DNA Isolation Micro Kit (Blue) and a leading competitor's kit (Red). Two µL of the DNA from each 200 µL of elution was used in a real-time PCR reaction (total reaction volume of 20 µL) with 5s rDNA primers. The real-time PCR was successful in amplifying 5s gene from both the rat and rabbit blood DNA, indicating that the DNA is of a high quality and can be used in sensitive downstream applications. Furthermore, Bio-Synthesis's-isolated DNA was amplified with a lower Ct value, indicating the higher yield and purity of DNA isolated using Bio-Synthesis's kit.
