BSI’s DNA Gel Extraction Kit enables rapid purification of DNA fragments that have been fractionated on agarose gels. The kit effectively removes gel contaminants, allows for buffer exchange and yields high quality DNA appropriate for many downstream applications.
Purification is based on spin column chromatography using BSI’s proprietary resin as the separation matrix. Breifly, the DNA of interest is run on an agarose gel and the band is then excised from the gel. Binding Solution is added to the gel slice to melt the agarose, and the released DNA is then bound to BSI’s column (BIND). Under these conditions only the DNA will bind to BSI’s resin while the agarose and other impurities are removed in the flowthrough. The bound DNA is then washed to remove any remaining impurities (WASH). Lastly, the purified DNA is eluted into 30 - 50 µL of the provided Elution Buffer or water (ELUTE). BSI’s DNA Gel Extraction Kit effectively removes gel contaminants, allows for buffer exchange and yields high quality DNA appropriate for many downstream applications. The kit is able to recover DNA ranging in size from 100 bp to 15,000 bp. Each column has a binding capacity of 25 mg, and the typical purification yield is up to 90% of the input amount.

The high yield of BSI ’s DNA Gel Extraction Kit is illustrated by purification of 1.5 µg of a 500 bp fragment from a 1% agarose gel using BSI ’s DNA Gel Extraction Kit and two other commercial kits. DNA recovery was quantified by running 300 ng (1/5th) of the input and 10 µL (1/5th) of the 50 µL DNA elution on a 1X TAE, 1% agarose gel followed by densitometry. The BSI kit shows a significantly higher efficiency compared to the competitor kits based on the average of three purifications.
