Controlling DNA functions using an external stimulus to quantitate at given moment can provide new tools for the analysis of analysis of the mechanisms underlying DNA recognition and DNA-mediated bioprocesses in cell biology and pharmacology. Photo-control modified oligonucleotide react with ultraviolet or visible light has a number of advantages over other external stimuli:

  • Light does not introduce contaminants into the reaction system,
  • Excitation wavelength can be controlled through the design of the photo-responsive molecule, and
  • It is now straightforward to control irradiation time and/or local excitation.

Asanuma's group has demonstrated when a photo-responsive modified oligo is directly attached to DNA as a receptor, photo-regulation of the bioprocess regulated by that DNA molecule could, in principle, be achieved. Such photo-responsive DNA could also be used as a switch in a DNA-based nano-machine1,2.

Bio-Synthesis offers several photo regulation modified base oligo synthesis. Contact Bio-Synthesis for oligo modification to block nuclease degradation.

All oligonucleotide synthesized at Bio-Synthesis is quality check by using either MALDI-TOF mass spectrometry, analytical HPLC,or polyacrylamide gel electrophoresis (PAGE) analysis. Final yields are determined using UV absorbance at OD260 In addition, we perform custom formulation and quality control tailored to meet your specific requirements.

Metalobase Modification Modification Code 5' Prime Internal 3' Prime
Azobenzene 300-400 nm (λmax = 330 nm) Y Y Y
4-Thiouridine 330-360 nm (λmax = 340 nm) Y Y Y
4-Thiothymidine 340-360 nm (λmax = 254 nm) Y Y Y
4-Thio-2'-deoxyuridine 330-360 nm (λmax = 340 nm) Y Y Y
6-Thio-2'-deoxyguanosine 340-360 nm (λmax = 254 nm) Y Y Y

Quality Control: Analytical HPLC, Gel eletrophoresis, and  MALDI-TOF Mass Spectrometry

Delivery times: 5-7 Working days

Packaging:    Dried

Shipping conditions: Room temperature

Storage conditions: -20 oC to -70 oC

Oligonucleotides are stable in solution at 4 oC for up to 2 weeks. Properly reconstituted material stored at -20 oC should be stable for at least 6 months. Dried DNA (when kept at -20 oC) in a nuclease-free environment should be stable for years.
