Bio-Synthesis offers reliable custom siRNA synthesis in ready-to-use duplex multi-scale quantities for your silencing applications. We offer a wide range of modifications and purification methods. Large-scale sterile, endotoxin-free preparations are available specifically for therapeutic applications. You may submit your siRNA sequence of choice, or use our design tool to configure a siRNA sequence. All custom siRNA oligonucleotides are manufactured under strict ISO standards and undergo QC by analytical HPLC, mass spectrometry, annealed in RNase-free water and duplex analyzed by analytical gel. Synthetic siRNA and shRNA constructs are just two of the numerous options available. Bio-Synthesis provides synthesis of sequence-specific custom siRNA for direct transfection, as well as synthesis of shRNA oligos, ~60 bases long purified DNA oligos to be cloned to form shRNA constructs. Our siRNAs (small interfering RNAs) are double-stranded RNA molecules, made of up to 30 RNA nucleotides with two nucleotides 3' overhangs. They are used to knock-down a specific gene of interest for identifying gene function, elucidating pathways and screening for potential new drug targets.

Custom Duplex siRNA Synthesis

Our siRNA oligos are available at a fixed price per duplex oligonucleotide strands of up to 30 nt target sequence from which both the sense and antisense strand will be synthesized, including 3' overhang bases. Contact us for longer oligos or spiked oligos which are available upon requests.

  • HPLC Purified:
    Please Note: The siRNA duplex is now dried down in the absence of buffer and the amount packaged per tube has changed, however, the total amount of material supplied remains the same.

    2. Dried duplexes are provided in the absence of universal buffer salts, unless otherwise specified upon ordering.
    3. The oligos are ready for use immediately upon resuspension.
    4. Typical shipping time is 6 business days for orders received before 3:00pm CST. Ship date will be confirmed upon order placement.
  • siRNA In-Vivo-Ready: A minimum of 90% purity. These duplexes are shipped deprotected with extra processing which also includes counter ion (Na+) exchange, dialysis, sterile filtration and endotoxin testing for the introduction of siRNAs into animals.

    Estimated shipping time is 10 business days for orders received before 3:00pm CST. Ship date will be confirmed upon order placement.

  • HPLC purified: In-Vivo-Ready custom synthesized siRNAs provide the highest quality siRNAs available anywhere with guaranteed purity of 97%. Pprocessing includes an extra level of HPLC purification, counter ion (Na+) exchange, dialysis to remove salt (to <1 microsiemens conductance at 50 µM siRNA), sterile filtration and endotoxin tested.

Modifications and Labelings

Custom siRNA is available with a variety of fluorescent labels and other modifications. Alexa Fluor dyes provide superior performance and can be directly substituted for other common dyes. Other fluorescent dyes available include fluorescein, rhodamine, Cy3, and Cy5. Custom siRNA may be labeled at either the 3' or the 5' end of the sense strand, and labels do not affect the biological activity of the siRNA. As well as fluorescent labels, many other backbone and base modifications are available including amino linkers, thio linkers, biotin, dabcyl and phosphate modifications. Note: all siRNA oligos are provided as lyophilized pellets to ensure stability during delivery.

Synthesis Platform

Bio-Synthesis provides siRNA by through proprietary synthesis and purification processes with final products that are ready-to-use. There is no need for further time-consuming and expensive HPLC or PAGE purification. The high-throughput synthesis and purification processes enable fast turnaround and high yields. Each duplex siRNA synthesis undergoes stringent quality control including MALDI-TOF mass spectrometry analysis that verifies identity of the component strands and analytical HPLC QC to verify purity of the duplex itself, a level of QC unparalleled in the industry. Our high performance grade siRNA is economically priced allowing the use of highly pure siRNA in all routine RNAi experiments. For very sensitive applications, custom synthesis of HPLC-purified siRNA is also available upon request.


Quality Control Options

Standard QC package

Analytical anion-exchange HPLC of oligonucleotide; purity guaranteed above 85% in case purification is required. TOF/MS for oligonucleotide identification.

Advanced QC package

Standard QC package and additionally:

Ion-pair reverse phase HPLC of the oligonucleotide at 60oC, purity guaranteed above 85% analytical reverse phase HPLC Endotoxin analysis of the oligonucleotide (*phosphorothioates may inhibit in the mechanism for endotoxin determinations Bio- Synthesis is using and therefore reading might be less accurate for these molecules) pH determination of a 1% oligonucleotide solution.

Custom QC Package:
