Gene targeting by modulation of DNA triplex stability using nucleobase modification have become an important potential therapetuic agent. These triplex forming oligonucleotides (TFOs) are a class of DNA oligonucleotides that are capable of biding to the major groove of duplex DNA forming a triplex helix. The formation of a triplex along the major groove competes with the binding of transcription factors and other proteins that are necessary for transcription. Triplex forming oligonucleotides such as bridged nucleic acids, BNAs provide sequence specificity and can be used to inhibit expression of specific genes. Furthermore TFOs can also be used as a diagnostic tool for identification of foreign DNA (bacterial or viral) or disease associated mutations.
