Cyclization of pepitdes has been shown to be a usefull approach in developing useful peptidic and peptidomimetic agents for diagnostic and therapeutic research. Since the structure of a peptide is determined by its sequence and the chemical environmnet in which it is placed, while the linear peptides are relatively unrestrained and are able to adopt a range of conformations that are not always ideal for some applications, cyclic peptide reduces the conformational freedom than of these flexible, linear molecules, and often results in higher receptor binding affinities by reducing unfavorable entropic effects1. Two strategies of cyclization used in our laboratory 1) Backbone to backbone cyclization strategies by the formation of the amide bond between the N-terminal and C-terminal amino acid residues, and 2) Cyclization of side chain amino acid such as:
 
Formation of disulfide bridges between cysteine, (homocycsteine)
The formation of lactam bridges between glutamic/aspartic acid and lysine residues
The formation of lactone and thiolactone bridges between amino acid containing carboxyl, hydroxyl or mercapto functional groups.
The formation of thioether or ether bridges between the amino acids containing hycroxyl or mercapto functional groups and other special methods.
