Using a new synthesis technology, Bio-Synthesis is able to meet our customer requirements for high quality DNA, RNA, 2'OMe, and 2'F-RNA,  BNA and base mimic analogs from mg to multi-gram quantity for pre-clinical, diagnostic or therapeutic studies. At Bio-Synthesis, DNA, RNA, BNA or any base analogs can be synthesized at mid-scale range from mg to multi-gram quantity and maintaining our quality commitment. All products are analyzed by PAGE, HPLC and mass spectroscopy. Turn around time varies based on sequence, purification and quantity requested. It is always advisable to order your material well in advance to ensure that you receive the material prior to establishing your studies.

Synthesis

Use of the dedicated equipment for large-scale synthesis enables synthesis of gram quantity oligonucleotide (under 25 mers) per one lot. All the products have their original lot numbers and are under the strict lot control.

Purification

Several types of purification methods

Standard Purification (by default)

Salt content is as important as purity in large-scale synthesis. This is the reason for setting RP-Cartridge and desalting as a default purification method for this service.

HPLC Purification

  • HPLC is well suited to purify any modified oligo or unmodified sequences up to 60 bases in length and is the method of choice for purify larger amounts of oligos (i.e. > 1 µmol). Purity of > 85% is guaranteed for oligo up to 40 bases 
  • Ion-exchange HPLC (IE-HPLC) is available to purify long oligos up to 60 bases. Anion-exchange resins separate oligos on the basis of charge.  The resolution of this technique is very high but their use is limited to purification of rather short oligos. For unmodified oligos between 11-40, purity of > 90% is guaranteed.
  • Dual HPLC Purification and Dual PAGE and HPLC Purification a combined purifications are applied for this service: HPLC + IEX HPLC purifications for 20 - 30mers oligos, and HPLC + PAGE purifications for 31 - 50mers oligos. Such combined purifications enable average of 98% purity by taking off most of base defective products or other impurities.
  • RNAase Free HPLC Purifications Originally developed for isolating RNA oligos, RNase-Free HPLC Purification has been extended to DNA oligos for use when there is a high sensitivity to ribonucleases. RNase-Free HPLC purification is carefully performed in an RNase-free environment; reagents, equipment and lab surfaces are monitored for RNase contamination.

In Vivo-Ready Purification

Bio-synthesis's In Vivo-Ready purification option provides high quality oligos that are ready for introduction into animals. Each oligo is HPLC purified, dialyzed to ensure that the oligonucleotide is salt free (conductance of <1 microsiemens for a 50 µM solution), sterile filtered, and endotoxin tested. Of course, all oligonucleotides are been analyzed by MALDI-TOF mass spectrometry and analytical HPLC, and sequencing.

Quality Control

Standard Quality Control

All synthesized DNA are subject to quality control by polyacrylamide gel electrophoresis. For the products under 35mer, any small deletion will be found by MALDI-TOF/MS spectrometry analysis system. RP-HPLC purification analysis is also applied for the purpose of complete QC.

Advanced Quality Control

We were among the first to adapt quality control by TOF/MS. Such experience enables us to offer some optional services such as primer sequencing (the direct analysis of primer sequence), or purity analysis data attachment by RP-HPLC or IEX-HPLC.

Pricing

Prices are for unmodified oligonucleotides between 15 and 25 bases. It includes purification and ethanol precipitation. Please request a quotation for other lengths or larger quantities. The backbone may consist of phosphorothioate or phosphodiester linkages, or a combination of the two. Prices may vary due to sequence composition.
