  N-terminal amino acid Nuclease degradation

N-Terminal

N Terminal sequencing of proteins and peptides provided as lyophilized samples, in solution or blotted on PVDF membrane as fee-for-service. Proteins and peptides can be sequenced five to ca. 40 steps depending on sample amount.
  Protein Sequencer Phosphate-diester DNA
4a Theory
The N-terminal amino acid reacts with phenylisothiocyanate (PITC). The derivatizing process results in a phenylthiohy dantoin (PTH) – amino acid. This amino acid is then sequentially removed while the rest of the peptide chain remains intact. Each derivatization process is a cycle. Each cycle removes a new amino acid.
  Picomole analyses Phosphorothioate linkages
4b Theory
Instrument on site is an Applied Bio system Precise Protein Sequencer - this instrument is well suited to high sensitivity low picomole analyses. Routinely used for the analysis of membrane bound gel electrophoresis separated proteins, HPLC separated tryptic digest fragments. Minimum of 12 picoMoles of a purified protein per run is required.
