Bio-Synthesis offers non-fluorescent labeled oligo synthesis and modification services using various types of modifiers. Example of using biotin labeled oligonucleotide in combination with streptavidin/avidin-conjugated fluorophores or enzymes (e.g. horseradish peroxidase), non fluorescent biotin-labeled DNA can be used for many applications such as Southern and Northern blotting, dot blots, DNA microarrays, immobilization of DNA.

We can directly conjugate biotins, non-fluorescent dark quenchers or other non-fluorescent modifiers to oligos at either end and/or internally at the time of synthesis using phosphoramidite chemistry or conjugate post-synthetically. Our non-fluorescent oligo conjugates are all HPLC or dual HPLC purified and quality checked by MALDI mass spectrometry.

Product 5' Internal 3' Function
Abasic Y Y Y dSpacer (abasic furan) is a tetrahydrofuran base, used to facilitate the determination of DNA structure by X-ray crystallography
Acridine Y Y N Acridine modified oligo are been used as an intercalating agent to target dsDNA and add stability to double helix.
Azobenzene Y Y Y Switch between cis and trans conformations an upon irradiation with UV or visible light for photo-control applications
Biotin Y Y Y Biotin-modified oligos bind tightly to streptavidin. The streptavidin can be labeled with fluorescent dyes and enzymes or mediate attachment to a solid surface to be used in a variety of molecular biology assays and purification methods.
Biotin Diol Linker Y Y Y Biotin contains a cleavable diol linker that can be used for detection, cleaved proteins that are not visible by means of streptavidine blot.
Biotin TEG Y Y Y Biotin-TEG a 15 atom mixed polarity triethylene glycol spacer which acts to minimize steric hindrance between biotin and the oligo, thereby providing streptavidin easy access to the biotin for capture and immobilization of the oligo.
dT-Biotin Y Y Y The internal version of this modification is attached to the oligo through a dT base. Incorporation of the internal version will add a dT nucleotide at that position.
Dual Biotin Y N N Two biotins are sequentially placed on the 5’-end, which increases the efficiency of streptavidin binding.
PC Biotin Y Y Y Photocleavable Biotin with optimal cleavage is obtained when exposure to long-wave UV light in the 300-350 nm spectral range. Cleavage releases the oligo with a 5’-phosphate group.
Cholesteryl TEG Y Y Y A photo reactive halogenated base, used to facilities the determination of DNA structure by X-ray crystallography
Desthiobiotin TEG Y Y Y Desthiobiotin, a biotin analog with affinity toward streptavidin biotin. Displacement of Desthiobiotin to streptavidin can occur by simply adding biotin to a reaction mixture.
DNP-TEG Y Y Y DNP with long triethylene glycol (TEG) spacer, commonly used to label oligonucleotides probes for use in hybridization applications
DNP-X Y Y Y DNP, a small molecule hapten having high immunogenicity
DOTA Y Y Y DOTA acts as a chelating agent and also possess a reactive site for covalent attachment to bio-targeting vectors such as antibodies, peptides and proteins. DOTA is widely used in nuclear medicine, MRI and optical imaging applications.
Psoralen C2 Y Y Y Psoralen is capable of intercalating between bases, and cross-link bases in both double-stranded and triple-stranded DNA
Psoralen C6 Y Y Y Psoralen is capable of intercalating between bases, and cross-link bases in both double-stranded and triple-stranded DNA
