Bio-Synthesis offers 2,4-dinitrophenyl DNP Oligonucleotide synthesis services. Oligonucleotides labelled with DNP (2,4-dinitrophenyl) can be used in analytical tests based on anti-DNP antibodies. This DNP with triethylene glycol (TEG) spacer can be introduced anywhere in the sequence and on either terminus.

DNP (2,4-dinitrophenyl) is small molecule hapten with high immunogenicity.  Antibodies raised against this haptens have higher affinities than other antibodies do for their targets,  making DNP particularly desirable as affinity tags for oligonucleotides1.

DNP (TEG) labeled oligonucleotides probes are often used in situ hybridization applications wth reduced background with subsequence antibody detection in contrast with biotinlylated probes2.  For example, in situ hybridization, Northern and Southern blotting2. After hybridization to their targets, these DNP-labeled probes are detected with anti-DNP antibodies that are labeled with dyes (for primary detection) or enzymes (for secondary detection using a fluorogenic, chemiluminogenic, or colorimetricsubstrate specific for the enzyme). To maximize signal obtained with such probes, Bio-synthesis recommends modifying the oligonucleotide probe with three DNP molecules, either grouped at the 5'-end or spaced about 10 bases apart 2.

In addition to the above straightforward anti-DNP antibody-based detection systems, oligo probes labeled with both a fluorescent dye and DNP also been used for highly-sensitive direct detection of antigens (at femtoMolar levels) in a rolling circle amplification (RCA)-based assay system 4.

Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at -20°C) in a nuclease-free environment should be stable for years.
