This is especially useful when immobilize the biotin-labelled DNA/RNA target sequence on the beads and incubate with the cell extract. Dual biotin labelled oligo keep biotinylated DNA/primers on the beads during heating after higher temperatures. Study have shown dual biotin prevents or effectively reduces leakage of biotinylated DNA from beads during heating1.
