Relative Quantitation of Gene Expression

The amplification of an endogenous control allows standardizing the amount of sample RNA or DNA added to a reaction. Researchers have used ß-actin, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), ribosomal RNA (rRNA), or other RNAs as an endogenous control to quantify gene expression.

Using the standard curve method or the comparative method, relative quantitation with data from the ABI P RISM® 7700 Sequence Detection System (using version 1.6 software) is possible.


The availability of different reporter dyes for the ABI PRISM 7700 Sequence Detection System makes it possible to amplify and detect the target amplicon and the endogenous control amplicon in the same tube (multiplex polymerase chain reaction [PCR]).
