Amplification of an endogenous control may be performed to standardize the amount of sample RNA or DNA added to a reaction. For the quantitation of gene expression, researchers have used ß-actin, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), ribosomal RNA (rRNA), or other RNAs as this endogenous control.
Relative quantitation with data from the ABI P RISM® 7700 Sequence Detection System (using version 1.6 software) can be performed using the standard curve method or the comparative method.
The availability of distinguishable reporter dyes for the ABI PRISM 7700 Sequence Detection System makes it possible to amplify and detect the target amplicon and the endogenous control amplicon in the same tube (multiplex polymerase chain reaction [PCR]).
