Post Purification Quality Control
Impurities such as salts, deletion peptides (created due to incomplete coupling), protecting groups and modified peptides can range from 10-50% by weight. It is recommended that all the peptides should be desalted or purified prior to use.
Samples containing peptide impurities are purified by reversed phase C-18 column (2.2 cm x 25 cm).
Multiple fractions are collected during the purification process
MALDI-TOF mass spectrometry and analytical HPLC on purified fractions
Fraction containing desired molecular weight and purity are pooled
MALDI-TOF and analytical HPLC is performed on the combined aliquot
