Alexa 647 Oligo Labeling

Alexa Fluor® 647 (AF647) is a premium far-red fluorescent dye designed for high-sensitivity fluorescence imaging and detection. It combines exceptional brightness, excellent quantum yield, and outstanding resistance to photobleaching, making it one of the most widely used fluorophores for advanced fluorescence microscopy, molecular diagnostics, and single-molecule imaging.

Alexa Fluor 647 is commonly conjugated to DNA, RNA, PNA, LNA, siRNA, antisense oligonucleotides (ASOs), aptamers, peptides, proteins, and other amino-modified biomolecules through NHS ester chemistry, producing a stable amide linkage with primary amino groups. Depending on the experimental design, the dye may also be introduced using click chemistry or other site-specific conjugation strategies.

The dye exhibits excitation and emission maxima of approximately 650 nm and 668 nm, respectively, making it compatible with 633 nm, 635 nm, and 640 nm laser systems commonly found on confocal microscopes, flow cytometers, fluorescence microscopes, and super-resolution imaging platforms. Alexa Fluor 647 provides excellent spectral separation from Alexa Fluor® 488, Alexa Fluor® 555, Alexa Fluor® 594, Cy3™, and FITC, making it particularly valuable for multicolor imaging and multiplex detection.

Because of its outstanding photophysical properties, Alexa Fluor 647 is one of the preferred fluorophores for single-molecule localization microscopy (SMLM), STORM, dSTORM, and other super-resolution imaging techniques.

Alexa Fluor 647 can be incorporated at the 5′ end, 3′ end, or internal amino-modified positions, and may be combined with numerous additional modifications including quenchers, biotin, thiol modifiers, amino modifiers, PEG linkers, click chemistry handles, GalNAc, cholesterol, peptides, lipid conjugates, and other custom chemistries.

Each labeled oligonucleotide is synthesized under stringent quality standards and is available with optional HPLC purification, together with analytical characterization by HPLC, LC-MS, MALDI-TOF, UV-Vis spectroscopy, and fluorescence analysis to verify labeling efficiency, sequence identity, and product purity.

Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at -20°C) in a nuclease-free environment should be stable for years.
