N3-Methyl-deoxy Thymidine (N3-Me-dT) is a N-alkyl pyrimidine. This base can be incorporate at any position within an oligonucletoide. The N3-Methyl group lower the hydrogen bonding with its complementary adenosine base during DNA replication. Studies also show that 3MeT is a strong block to the Klenow fragment of DNA polymerase I also. N3-Methyl dT modified oligonucletoides are been used in the binding study of antisense oligos to their mRNA targets and in the enzymatic mechanisms in the repair of DNA alkylation or UV-based DNA damage.
