Synthetic siRNA and shRNA constructs are two of the numerous options available. BSI provides the custom synthesis of sequence-specific siRNA for direct transfection, as well as the routine synthesis of shRNA oligos, ~60 bases long purified DNA oligos to be cloned to form shRNA constructs . Our siRNAs (small interfering RNAs) are double-stranded RNA molecules, made of 19 RNA nucleotides with two nucleotides (usually DNA bases) 3' overhangs. They are used to knockdown a specific gene of interest for identifying gene function, elucidating pathways and screening for potential new drug targets.
Custom siRNA is available with a variety of fluorescent labels and other modifications. Alexa Fluor dyes provide superior performance and can be directly substituted for other common dyes. Other fluorescent dyes available include fluorescein, rhodamine, Cy3, and Cy5. Custom siRNA may be labeled at either the 3' or the 5' end of the sense strand, and labels do not affect the biological activity of the siRNA. As well as fluorescent labels, many other backbone and base modifications are available including amino linkers, thio linkers, biotin, dabcyl and phosphate modifications. All siRNA oligos are provided as a lyophilized pellet to ensure stability during delivery.
