Xtremer Single-stranded DNA (ssDNA) fragments are important research tools in molecular biology. ssDNA can be used in diagnostic assays, CRISPR gene therapy, DNA nanotechnology and many other applications. ssDNA oligonucleotides are typically produced via well established chemical solid-phase DNA synthesis on polymeric supports. One advantage is that almost any type of nucleotide analog can be introduced at well defined positions. However, as the length is extended, the expected yields will diminish somewhat. To counter the diminished yield, Bio-Synthesis has developed enzymatic synthesis methods, allowing us to produce single-strand DNA reaching as much as 10,000 nt in some cases, while 3000-5000 nt is common.

DNA synthesis by enzymatic method is a powerful alternative to chemical DNA oligo synthesis and is useful when long DNA (>400 nt) is needed. Yields of chemically synthesized DNA decrease dramatically with increasing length. Thus with current chemistries, it is not efficient to synthesize long, high quality DNA chemically. We have therefore developed a unique enzymatic DNA synthesis method to produce long single stranded DNA (ssDNA) for lengths in the thousands of bases which are often used as repair templates in gene knockin experiments, in vitro transcription, and more.
