Alkaline Phosphatase AP Oligonucleotide Conjugate

Synthetic oligo probe can be functionalized any position and covalently cross-linked to alkaline phsophatase using homobifunctional cross-linkers. Modified oligonucleotides are purified prior conjugation with alkaline phosphatase (AP). After conjugation, final product are further purified with HPLC. Final concentration are determine by UV spectrum of AP (280 nm) and oligo (260 nm) and analyze by gel electrophoresis of oligo-HRP conjugates. The delivered AP-oligonucleotide conjugate from Bio-Synthesis, contains less than 5-10 % enzyme oligonucleotide dimer complex.
When designing oligonucleotide for AP conjugation, please avoid sequence containing palindromes or self complementary sequence. Alkaline phosphatase modified oligo is only available with HPLC 1 umole synthesis scale. A long PEG crosslinker myabe used for the flixibility and preservation of Alkaline phosphatase activity for longer oligonucleotide.
