Bio-Synthesis offers Spacer 9 PEG oligonucleotide modification at various scales. PEG Spacer 9 is a triethylene glycol chain 9 atoms long (6 carbons and 3 oxygens). Spacer 9 can be incorporated either at the 5'-end or 3'-end of an oligo or internally. Multiple insertions can be used to create long spacer arms.

Spacer 9 has been used for a number of different applications. It can form a non-nucleotide bridges in hairpin loops of an oligonucleotides,1 link oligonucleotides to epitopes for drug development,2 and for solid-phase immobilization of hybridization probes.3 Multiple incorporations of Spacer 9 have been used to form long, flexible linker arms between the two domains (double-helix forming and triple-helix forming, respectively) of a bifunctional DNA oligonucleotide, in order to maximize the binding flexibility of the two domains for their respective targets.4 This Spacer 9 modified oligonucleotide have been used to form a peptide nucleic acid PNA-DNA conjugate for use in site-directed recombination applications,1,2 but can be used for any oligonucleotide-based application requiring a long spacer arm.

Purification:

Spacer modified oligonucleotide can be accomplished by various methods. Desalting or cartridge purification is acceptable, however, additional purification by HPLC is strongly recommended.
