SMCC-maleimide (succinimidyl 4-[N-maleimidomethyl] cyclohexane-1-carboxylate) is a heterobifunctional cross-linker that can be covalently labeled at any position within an oligonucleotide that contains a primary amine. The succinimidyl ester group of SMCC-maleimide reacts with the primary amine to form a stable amide bond, which serves as a connector or linker to attach the oligonucleotide to other thiol-containing molecules, such as peptides or proteins. SMCC-maleimide oligonucleotide conjugates are a versatile tool in bioconjugation chemistry.

SMCC-maleimide is modified post-synthetically, meaning that the oligonucleotide must first be synthesized and then modified with SMCC-maleimide. The precursor amino-modified oligonucleotide is recommended to be purified by HPLC prior to labeling.
