Oligo Modifications for Cell Delivery and Uptake

Bio-Synthesis provides a collection of oligonucleotides modified with a multitude of lipophilic compounds including vitamins. These modified oligonucleotides are useful for various therapeutic applications. Unmodified oligonucleotides are hydrophilic in nature and dissolve well in water or various buffers. However, modification of oligonucleotides with lipophilic molecules or vitamins changes their solubility and make oligonucleotides more amphiphilic enhancing their transport through lipid membranes needed for a cellular update. Furthermore, oligonucleotides containing lipophilic compounds enable a variety of therapeutic approaches such as antisense technologies, siRNA, aptamer and immunostimulatory methods as well as drug delivery.

Lipophilic oligomodification enables powerful solutions in antisense application, siRNA, aptamer and immunostimulatory research and drug delivery. Contact Bio-Synthesis for lipophilic modified oligonucleotide synthesis service.

Lipophilic Oligonucleotide Modifications

Name Postion within Properties and Applications
Cholesterol cell1 Enhanced delivery and uptake in DNA antisense, siRNA or for artificial DNA-nanostructure.
α-Tocopherol cell2 Known as vitamin E. Used as lipophilic carrier for siRNA..
Trolox cell3 A derivative of vitamin E (tocopherol). It is more hydrophilic and has antioxidant properties.
Palmitate cell4 Fatty acid modified oligonucleotide. Conjugates guide protein targeting and improve cell delivery.
Stearate cell5 Fatty acids modified oligonucleotides can be used as alternative lipophilic carrier.

Vitamin Oligonucleotide Modifications

Name Postion within Properties and Applications
5'-Niacin cell6 An essential vitamin, known for its therapeutic effects as a lipid lowering drug.
bisDMT-Pyridoxine cell7 Known as vitamin B6.

Quality Control:

Analytical HPLC, Gel eletrophoresis, and  MALDI-TOF Mass Spectrometry

Delivery times:

5-7 Working days

Shipping conditions:

Room temperature

Storage conditions:

-20 oC to -70 oC

Oligonucleotides are stable in solution at 4 oC for up to 2 weeks. Properly reconstituted material stored at -20 oC should be stable for at least 6 months. Dried DNA (when kept at -20 oC) in a nuclease-free environment should be stable for years.
