Propargyl dU contains an alkyne group that can be introduced to an oligonucleotide at time of synthesis using phosphoramidite chemistry. This modification can be introduced internally or at the 5' terminus. The Progargyloxy dU oligo can be conjugated with azide bearing agents and involves copper-catalyzed azide/alkyne cycloaddition reaction. Ligation of oligonucleotides containing 5' azide with 3' propargy group using click chemistry to obtain long DNA construct, has been used as PCR template. This linkage has shown to be compatible transcription and rolling circle amplification, as well as gene expression in E. coli. Long RNA containing triazole linkage has shown to retain its activity at the substrate cleavage site.
