Gene targeting by modulation of DNA triplex stability using nucleobase modification have become an important potential therapetuic agent. These triplex forming oligonucleotides (TFOs) are a class of DNA oligonucleotides that are capable of biding to the major groove of duplex DNA forming a triplex helix. The formation of a triplex along the major groove competes with the binding of transcription factors and other proteins that are necessary for transcription. Triplex forming oligonucleotides such as bridged nucleic acids, BNAs provide sequence specificity and can be used to inhibit expression of specific genes. Furthermore TFOs can also be used as a diagnostic tool for identification of foreign DNA (bacterial or viral) or disease associated mutations. Feature of using Triplex forming nucleobase modified oligonucleotide

Feature and Benefits

  • Useful as a therapeutic Agents
  • Useful as a diagnostic for infectious diseases or genetic mutations
  • Nucleosides can be assembled into defined oligomers to target specific genes

Bio-Synthesis offers several triplex forming modified nucleobases. These modified bases are capable of forming hydrogen bonds with interstrand purine-pyrmidine base pairs and can be assembled into defined oligomers that have the ability to bind to specific sequences of DNA and allowing them to trageted any known DNA or RNA sequences. Contact Bio-Synthesis for triplex forming oligonucleotide synthesis.

All oligonucleotide synthesized at Bio-Synthesis is quality check by using either MALDI-TOF mass spectrometry, analytical HPLC,or polyacrylamide gel electrophoresis (PAGE) analysis. Final yields are determined using UV absorbance at OD260 In addition, we perform custom formulation and quality control tailored to meet your specific requirements.

Quality Control: Analytical HPLC, Gel eletrophoresis, and  MALDI-TOF Mass Spectrometry

Delivery times: 5-7 Working days

Packaging:    Dried

Shipping conditions: Room temperature

Storage conditions: -20 oC to -70 oC

Oligonucleotides are stable in solution at 4 oC for up to 2 weeks. Properly reconstituted material stored at -20 oC should be stable for at least 6 months. Dried DNA (when kept at -20 oC) in a nuclease-free environment should be stable for years.
