Custom peptides can be synthesized in a variety ways for a multitude of purposes
in order to produce research-grade or bulk API polypeptides. These peptides can
also be modified during synthesis for specific diagnostic and therapeutic applications.
Liquid Phase Custom Peptide Synthesis (LPPS) is used for very short
dipeptide synthesis, while large scale Solid Phase Custom Peptide Synthesis
(SPPS) is suited for preparing biologically active peptides including long, difficult
and hydrophobic peptides.
Bio-Synthesis offers competitive prices, world-class support and fast delivery time.
Our ISO9001 quality process ensures that each peptide is synthesized to strict quality
Overnight service is available for crude, unmodified peptide < 10 residues with
amorphous product. An express service fee may apply. Regular turnaround time is
3-7 days for regular peptides. Additional time will be needed for special peptides
and purification needs.
While peptides with normal difficulty have maintained a standard pricing, Bio-Synthesis
offers an obligation-free consultative quote on all peptide sequences. Bio-Synthesis
will also assist you in selecting the most appropriate peptide sequence, purity
and quantity for your needs by using commercial software. Based on our own internal
database to synthesize sequences, a determination is made regarding the complexity
of the synthesis. This includes a hydrophobicity/hydrophilicity plot (Hoop-Woods
and Kyte-Doolittle algorithms). For antibody production we also include antigenic
index (Jameson-Wolf) and surface probability evaluations. In many cases, feedback
provided to you on the feasibility of the candidate sequences can result in a better
choice of a peptide which is more stable, more soluble or more economical. After
the synthesis feasibility is assessed, our sales team will provide a quote from
which you can choose your preferred specifications.
To request a quote, a peptide assessment, or to order, contact us by email,
fax, phone, or through the user friendly quotation request form on this website.
Naturally we will need full information on sequences, amount and desired purity
level(s), which in many instances will be finalized with you after a technical discussion
with our experienced technical representatives. All sequence information will be
kept strictly confidential.
Peptide synthesis is carried out by FMOC chemistry under continuous flow conditions
using PEG-Polystyrene resins. At the completion of synthesis, peptides are cleaved
from the resin and de-protected using TFA/DTT/H2O/Triisopropyl saline cleavage cocktail.
Peptides are then precipitated from the cleavage cocktail using cold diethyl ether.
The precipitate is then washed three times with the cold ether and then dissolved
in a crude buffer containing H2O/ACN/HOAC (75/20/5) prior to lyophilization. QC
is based on an analytical-scale reverse-phase HPLC chromatogram and the mass analysis
by MALDI will be provided for every peptide to ensure the highest quality of synthetic
All peptides are analyzed by MALDI Mass Spectrometry to determine the molecular
weight of the synthesized peptide, free of charge. Comparison of the theoretical
molecular weight with the experimentally determined molecular weight confirms the
absence of amino acid deletions or double couplings. MALDI is an excellent method
for determining the presence of the desired product, but an inadequate method for
the determination of purity. Due to the variant efficiencies of peptide ionization
in the MALDI instrument, peak intensities in MALDI spectra are not necessarily quantitative.
For this reason all non-crude peptides at Bio-Synthesis are analyzed by Reverse-Phase
HPLC (RP-HPLC) to determining the purity. Additional Q.C. qualification is also
available such as amino acid analysis, NMR, CE, as well as peptide content determination
to confirm the identity and demonstrate the high quality of our custom peptides.
Successful synthesis depends on the sequence of the amino acids. Likewise, the coupling
efficiency of amino acids in the sequence and ultimate purity of the synthesized
product is also related to the sequence. Impurities such as salts, deletion peptides
(created due to incomplete coupling), protecting groups and modified peptides can
range from 10-50%.
Purification of synthetic peptides is carried out by HPLC using reverse phase C4
and C18 columns. Alternative purification methods (e.g., ion exchange and gel filtration
chromatography) are used when needed. For purity requirements of >95%, Bio-Synthesis
runs its HPLC analyses at a gradient of 0-60% over 45 minutes. Like a close-up
photograph, this insures that peptide validation reveals even the smallest of impurities.
Peptides that meet or exceed the purity requirements are checked again by MALDI
or additional extended quality control or preparative services.
Each peptide is shipped with a Certificate of Analysis based on up-to-date analysis
of the peptide. The Certificate of Analysis includes details of the peptide, the
quantity and purity supplied, and helpful information on the solubility of the peptide
in the buffers used during purification.
All purified peptides also include a printout of the analytical HPLC profile and
electrospray MS data. Peptides are also shipped with ‘A guide to handling peptides’
and ‘Interpreting the results of peptide analysis’
BSI's peptides are packed under dry argon gas to minimize oxidation or other types
of degradation during shipment and storage. After opening, we recommend peptides
are stored dry in a freezer (details given in our technical guide to handling peptides

Peptides can be synthesized from immunograde purity (> 70 %, sufficient for generating
or testing antibodies) to higher purity (+ 95 %, required in enzymological and biological
activity studies and other applications). Peptides can be synthesized up to 200
amino acids and longer peptides can be made through recombinant peptide technology.

Synthetic polypeptides have spurred the development of different applications in
biochemistry, molecular biology, immunology, and medicine. Synthetic peptides may
be useful in epitope-specific antibodies against pathogenic proteins, the study
of protein functions and the identification and characterization of proteins. Furthermore,
synthetic peptides are used to study enzyme-substrate interactions within important
enzyme classes such as kinases and proteases, which play a crucial role in cell
In diagnostics, a set of homologous synthetic peptide binding ligands would be useful
directing reagents to particular epitopes in a protein, the detection of peptide
hormones, and many other applications. In therapeutic applications, peptide drugs
are either resembled as naturally-occurring peptides or altered natural peptides.
If the analogue is more biochemically active than the original peptide it is known
as an agonist and if it has the reverse effect is known as an antagonist.
Synthetic peptides are used as standards and reagents in mass spectrometry (MS)-based
applications. Synthetic peptides play a central role in MS-based discovery, characterization
and quantitation of proteins, especially those that serve as early biomarkers for
We constantly seek ways to improve our quality systems and welcome any suggestions
