Bio-Synthesis fluorescein isothiocyanate, FITC oligonucleotide services. 6-FITC is a bright green fluorophore that is sensitive to changes in pH and can photobleach rapidly unless an antifade agent is used. FITC has excitation max of 490 nm and emission max of 525. 6-FAM can be use to replace FITC. This dye can be directly incorporate at time of synthesis. FITC dye has two mixture of isomers at 5 or 6 position of the ring. Single isomer incorporation is also avaiable. FITC dye can be incorporated at any position within an Oligonucleotide. A primary amine modified oligonucleotide is used to covalently attach with dye via NHS conjugation chemistry; other cross-linking chemistries are also available depending on particular project specifications. Dual HPLC purification is used to remove failure sequences during first HPLC purification, after labeling, the second HPLC is performed to remove unlabeled oligonucleotides and excessive dye in order to obtain a full length labeled oligonucleotide.
