What Type of RNA Synthesis Services do You Provide?

Bio-Synthesis provides two types of RNA synthesis services:
1. Chemical RNA synthesis
2. Enzymatic RNA synthesis

Chemical RNA Synthesis
Using phosphoramidite chemistry, chemical RNA synthesis method ensures the exact sequence as requested, specialized chemical modifications, bases and labeling could also be incorporated at specific location. However chemical method has it limitation (up to 150-300 in length depending on sequence composition). chemical method also has a very low yield, so the overall cost is higher than the alternative method. RNA synthesis using chemical method deliver as 5' and 3' OH.


Enzymatic RNA Synthesis (IVT)
IVT allows for very long sequences (up to 10,000) to be generated and is easier to scale up. The advantage of IVT is that a sequences may have three added bases leading the sequence as a result generated by the promoter.

Long RNA synthesis is generated using bacteriophage polymerase such as T7, T3 or SP6. Depending on the complexity and length of the sequence, RNA generated using polymerase T7 and T3, could generate an additional GGG, and SP6 could generate an additional GAA as a result of the promoter sequence. For most molecular biology applications, the three extra bases on the 5' end should not be a problem, considering the much greater length of entire RNA sequence.
