Are there any interference from polysorbate 20 for hydrolysis protein-AAA test?

  1. The polysobate should not interfere. Since the protein is quite diluted we will need to concentrate it by lyophylization prior to hydrolysis.
  2. Alternatively, the concentration could be done using a polysorbate free buffer such as a 10 mM sodium borate buffer and a spin down concentrator prior to hydrolysis.
  3. Or, using a protein precipitation method such as the chloroform/methanol/water method prior to hydrolysis.
