Do you test for Glutamine in animal serum?
This could be done as follows:
- An aliquot of the serum will need to incubated with an aliquot of sodium borate buffer over night at pH 8.3 to dissolve the glutamine quantitatively.
- Sulphosalicylic acid will need to be added to keep the amino acids stable
- The proteins in the serum will need to be separated, e.g. by ethanol precipitation or a similar precipitation
- The glutamine in the supernatant will be analyzed
- An internal standard such as norleucine will need to be used to spike the sample for normalization of the final data
- We most likely will need to outsource the amino acid analysis to perform a ninhydrin based analysis
- We can do the extraction
