Amyloid Beta Peptides (Aβ)

Definition

Amyloid beta (Aβ) peptides are aggregation-prone peptides derived from proteolytic cleavage of the amyloid precursor protein (APP). These peptides self-assemble into β-sheet–rich fibrillar structures that form the core of amyloid deposits observed in neurodegenerative diseases, particularly Alzheimer’s disease (AD).

Amyloid fibrils are filamentous protein aggregates ranging from nanometers to microns in length and are characterized by highly ordered cross-β sheet architecture composed of parallel or antiparallel β-strand alignments.

Structural Characteristics

Solid-state NMR (SSNMR) studies, combined with computational energy minimization, have provided detailed structural models of Aβ fibrils, particularly Aβ1–40 under physiological conditions (pH 7.4, 24°C).

• Two β-strands per Aβ1–40 molecule spanning residues ~12–24 and 30–40
• A connecting loop region (residues 25–29)
• Parallel, in-register stacking of Aβ molecules along the fibril axis
• Protofilaments composed of four β-sheets separated by ~10 Å

These structural characteristics form the basis for aggregation assays, fibrillization studies, and inhibitor screening applications.

Role in Alzheimer’s Disease

Alzheimer’s disease is defined at the molecular level by the accumulation of amyloid deposits and neurofibrillary tangles. Aβ peptides:

• Are principal constituents of extracellular amyloid plaques
• Are derived from APP cleavage by β- and γ-secretases
• Exist in multiple aggregation states including monomers, soluble oligomers, protofibrils, and mature fibrils

Among these forms, soluble oligomers are widely considered the most neurotoxic species.

Mechanisms of Toxicity

Calcium Dysregulation and Membrane Disruption

Oligomeric Aβ42 has been shown to rapidly elevate intracellular Ca²⁺, increase membrane permeability, and induce unregulated ionic flux that disrupts cellular homeostasis. Monomeric and fibrillar forms do not exhibit the same acute calcium-disruptive effects. Evidence suggests toxicity arises from generalized membrane permeabilization rather than activation of endogenous calcium channels.

This mechanism is considered a shared feature across multiple amyloidogenic proteins.

Related Amyloidogenic Peptides

Amyloidogenic aggregation is not limited to Aβ. Other disease-associated peptides include:

• Islet amyloid polypeptide (IAPP, amylin) in type 2 diabetes
• Prion protein fragments
• Polyglutamine-expanded proteins
• Lysozyme variants

In type 2 diabetes, intracellular oligomers of IAPP are believed to contribute to pancreatic β-cell loss via mechanisms similar to Aβ oligomer toxicity.
