Unmodified PNA

Refer to PNa’s with no labels or fluorophores attached to it, but they have the N termini free or blocked. For these PNA’s these are the options:

  Unmodified PNA Unmodified PNA with Options*

Free Amine
The PNA molecule contains a free primary amine at the N terminus. This free primary amine permits users to introduce modifications.

     

Addition of Lysine
Add one or more Lysines to either the the N or C-terminus of your PNA probe. Adding a Lysine introduces a positive charge into the PNA and increases water solubility and cellular uptake.

Acetylated:
The PNA molecule contain an acetyl group at the N-terminus. This blocks any further reactivity at the N terminus.

  Addition of Solubility Enhancer
Solubility enhancer which do not bring any charge to molecular but enhances solubility nevertheless. Can be added at the C or N terminus.

Alkylation
The PNA molecule contains an alkyl group at the N terminus.

  Linker Addition
O-Linker can also be used to increase the solubility of PNA in water. Normally, linkers are added to the N-terminus, where a user can attach its own label.

*In order to increase the solubility of your PNA and to avoid aggregation, we suggest adding one or more Lysines at the N terminus, the C terminus or both if the particular application so calls for.

 PNA Modifications

For target detection, in situ detection or other applications, we can attach your PNA molecule with a variety of fluorophores including but not limited to the list below. BSI continually developing the incorporation of labelings and modifications. If a modifications or labelings not listed below, please inquire. It may be available as a product under development or we may be able to ad it to our list of R&D projects.

  Linkage at the N-Terminus, via O-linker
