Custom PNA Synthesis Overview

Synthesis of peptide nucleic acids (PNA's) waholm, Berg, and Buchardt in 1991, a new, completely artificial DNA/RNA analog by replacing sugar phosphate backbons was introduced by Nielsen, Ege of natural ncueic acid with syntheic peptide backbone.

Unlike DNA or DNA analogs, thses peptide-like DNA structures have the ability to form extremly stable complexes with complementary DNA oligomers and resistance to hydrolytic reactions and, thus, not expected to be degraded inside a living cell.

The impressive property of PNA's are their ability to form extremely stable complexes with complementary DNA oligomers.

This stability suggests PNA's are superior reagents in antisense and antigene applications, and creates other uses for which sequence specific but thermally stable complexes are required.
