Acrydite can be place placed at the 5' end of an oligonucleotide. The acrylamide group of acrydite modified oligo is covalently react with thiol-modified surface or can be incorporated into polyacrylamide gels during polymerization via a thioether bond. The immobilization leaves the oligonucleotides available for hybridization. The Acrydite modification is insensitive to heat and other denaturing agents. Acrydite-modified oligos are stable in aqueous solutions over a wide range of temperature and pH.
