DY-350XL UV MegaStokes® Fluorescent Dye Oligonucleotide Modification

DY-350XL UV MegaStokes® is an advanced fluorescent dye developed by Dyomics GmbH as part of its UV MegaStokes® fluorophore family. Unlike conventional fluorophores that exhibit relatively small differences between excitation and emission wavelengths, UV MegaStokes® dyes are specifically engineered with an exceptionally large Stokes shift, allowing efficient excitation with ultraviolet light while emitting fluorescence at significantly longer wavelengths.

The large spectral separation dramatically reduces excitation light bleed-through into the emission channel, minimizing background fluorescence and improving signal-to-noise ratios. These properties simplify optical filter selection and make DY-350XL UV MegaStokes® particularly valuable for high-sensitivity fluorescence detection, quantitative imaging, and multiplex fluorescence assays.

Bio-Synthesis offers custom DY-350XL UV MegaStokes®-modified oligonucleotides for DNA, RNA, PNA, LNA, siRNA, antisense oligonucleotides (ASOs), aptamers, PCR probes, molecular beacons, FISH probes, CRISPR guide RNAs, and numerous custom nucleic acid constructs. The dye may be incorporated at the 5′ terminus, 3′ terminus, or internally through amino-modified nucleotides using appropriate linker chemistries.

The dye incorporates Dyomics' hydrophilic molecular design to improve aqueous solubility while minimizing nonspecific adsorption to biological samples and laboratory surfaces. Combined with excellent photostability, DY-350XL UV MegaStokes® supports prolonged fluorescence imaging and repeated excitation without significant photobleaching, making it well suited for demanding fluorescence microscopy and analytical applications.

Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at 20°C) in a nuclease-free environment should be stable for years.
