Dyomic DY-431, Green Emission

DY-431 is a high-performance fluorescent dye developed by Dyomics GmbH for advanced fluorescence labeling applications. As a member of the Dyomics® DY Series, DY-431 combines high fluorescence intensity, excellent photostability, and superior aqueous solubility to provide reliable labeling of oligonucleotides and other biomolecules for research and diagnostic assay development.

Designed for excitation by violet laser sources, DY-431 produces bright blue fluorescence with excellent resistance to photobleaching. Its hydrophilic molecular design minimizes nonspecific adsorption while improving labeling efficiency and fluorescence reproducibility. These characteristics make DY-431 particularly useful for high-sensitivity fluorescence microscopy, confocal imaging, flow cytometry, fluorescence in situ hybridization (FISH), and multiplex detection systems.

Bio-Synthesis offers custom DY-431-modified oligonucleotides for a broad range of nucleic acid formats, including:

  • DNA oligonucleotides
  • RNA oligonucleotides
  • siRNA
  • Antisense oligonucleotides (ASOs)
  • PNA
  • LNA/BNA oligonucleotides
  • Aptamers
  • PCR probes
  • Hybridization probes
  • Molecular Beacon probes
  • FISH probes
  • CRISPR guide RNAs

DY-431 may be incorporated at the 5′ terminus, 3′ terminus, or internally through amino-modified nucleotides using suitable linker chemistries.

The dye is compatible with numerous additional Bio-Synthesis modifications, including:

  • Biotin
  • Digoxigenin
  • Amino modifiers
  • Thiol modifiers
  • PEG linkers
  • Azide
  • DBCO
  • BCN
  • Tetrazine
  • Cholesterol
  • GalNAc
  • Peptide conjugates
  • Lipid conjugates
  • Fluorescent quenchers

Its spectral properties allow DY-431 to function effectively in multiplex fluorescence experiments together with green-, yellow-, orange-, red-, and near-infrared fluorophores. The dye's excellent photostability enables prolonged imaging sessions while maintaining consistent fluorescence intensity.

Typical research applications include:

  • DNA and RNA fluorescent labeling
  • Fluorescence microscopy
  • Confocal microscopy
  • Flow cytometry
  • Fluorescence in situ hybridization (FISH)
  • Multiplex fluorescence imaging
  • Quantitative PCR probes
  • Molecular Beacon assays
  • Hybridization assays
  • Aptamer imaging
  • DNA nanotechnology
  • Biosensor development

Every DY-431-modified oligonucleotide is synthesized using automated solid-phase phosphoramidite chemistry and is available with optional HPLC or PAGE purification. Comprehensive analytical quality control includes HPLC, LC-MS, MALDI-TOF, UV-Vis spectroscopy, and fluorescence verification to confirm sequence integrity, purity, and successful dye incorporation.
