Dyomic DY-505, Green Emission

DY-505 is a premium fluorophore developed by Dyomics GmbH for advanced fluorescence labeling of nucleic acids and other biomolecules. As a member of the Dyomics® DY Series, DY-505 combines excellent brightness, high quantum yield, superior resistance to photobleaching, and low nonspecific binding to deliver reliable fluorescence performance in demanding molecular biology applications.

The dye is efficiently excited using the widely available 488 nm blue laser, making it fully compatible with modern fluorescence microscopes, confocal imaging systems, flow cytometers, laser scanners, fluorescence plate readers, and imaging instruments. Its bright green fluorescence provides excellent sensitivity while supporting multiplex detection together with orange-, red-, and near-infrared fluorophores.

Bio-Synthesis offers custom DY-505-modified oligonucleotides for numerous nucleic acid formats, including:

  • DNA oligonucleotides
  • RNA oligonucleotides
  • siRNA
  • Antisense oligonucleotides (ASOs)
  • PNA
  • LNA/BNA oligonucleotides
  • Aptamers
  • Hydrolysis (TaqMan®) probes
  • Molecular Beacons
  • Hybridization probes
  • FISH probes
  • CRISPR guide RNAs

DY-505 may be incorporated at the 5′ terminus, 3′ terminus, or internally using amino-modified nucleotides and appropriate linker chemistries.

The dye is compatible with numerous Bio-Synthesis modifications, including:

  • Biotin
  • Digoxigenin
  • Amino modifiers
  • Thiol modifiers
  • PEG linkers
  • Azide
  • DBCO
  • BCN
  • Tetrazine
  • Cholesterol
  • GalNAc
  • Peptide conjugates
  • Lipid conjugates
  • Fluorescent quenchers

DY-505 exhibits excellent resistance to photobleaching, making it suitable for prolonged fluorescence microscopy, repeated laser scanning, live-cell imaging, and quantitative fluorescence measurements. Compared with traditional fluorescein-based dyes, DY-505 generally offers improved photostability and fluorescence performance while maintaining compatibility with standard FITC optical filter sets and 488 nm excitation sources.

Typical research applications include:

  • DNA and RNA fluorescent labeling
  • Fluorescence microscopy
  • Confocal microscopy
  • Flow cytometry
  • Fluorescence in situ hybridization (FISH)
  • Quantitative PCR probes
  • Molecular Beacon assays
  • Hybridization probes
  • Live-cell fluorescence imaging
  • Aptamer imaging
  • DNA nanotechnology
  • Biosensor development
  • Multiplex fluorescence detection

Every DY-505-modified oligonucleotide is synthesized using automated solid-phase phosphoramidite chemistry and is available with optional HPLC or PAGE purification. Comprehensive analytical quality control includes HPLC, LC-MS, MALDI-TOF, UV-Vis spectroscopy, and fluorescence verification to confirm sequence identity, purity, and successful dye incorporation.
