Dyomic DY-549P1, Orange Emission

DY-549P1 is a premium fluorophore developed by Dyomics GmbH for advanced fluorescence labeling of nucleic acids and biomolecules. As a specialized derivative within the DY-549 dye family, DY-549P1 combines high quantum yield, excellent photostability, and low nonspecific binding with excellent aqueous solubility, providing bright and reproducible fluorescence signals in demanding molecular biology applications.

The dye is optimized for excitation using 532 nm DPSS lasers, 543 nm HeNe lasers, and 561 nm solid-state lasers, making it compatible with most modern fluorescence microscopes, confocal imaging systems, flow cytometers, laser scanners, and fluorescence imaging instruments. Its bright orange emission provides excellent spectral separation from green and far-red fluorophores, making DY-549P1 particularly valuable for multiplex fluorescence experiments.

Bio-Synthesis offers custom DY-549P1-modified oligonucleotides for numerous nucleic acid formats, including:

  • DNA oligonucleotides
  • RNA oligonucleotides
  • siRNA
  • Antisense oligonucleotides (ASOs)
  • PNA
  • LNA/BNA oligonucleotides
  • Aptamers
  • Hydrolysis (TaqMan®) probes
  • Molecular Beacons
  • Hybridization probes
  • FISH probes
  • CRISPR guide RNAs

DY-549P1 may be incorporated at the 5′ terminus, 3′ terminus, or internally through amino-modified nucleotides using appropriate linker chemistries.

The dye is compatible with numerous Bio-Synthesis oligonucleotide modifications, including:

  • Biotin
  • Digoxigenin
  • Amino modifiers
  • Thiol modifiers
  • PEG linkers
  • Azide
  • DBCO
  • BCN
  • Tetrazine
  • Cholesterol
  • GalNAc
  • Peptide conjugates

  • Fluorescent quenchers

DY-549P1 exhibits excellent resistance to photobleaching and maintains stable fluorescence during prolonged illumination, making it suitable for quantitative fluorescence microscopy, repeated laser scanning, live-cell imaging, and high-sensitivity molecular detection. Its spectral profile enables efficient multiplexing with blue-, green-, red-, and near-infrared fluorophores.

Typical research applications include:

  • DNA and RNA fluorescent labeling
  • Fluorescence microscopy
  • Confocal microscopy
  • Flow cytometry
  • Fluorescence in situ hybridization (FISH)
  • Multiplex fluorescence imaging
  • Quantitative PCR probes
  • Molecular Beacon assays
  • Hybridization assays
  • Live-cell fluorescence imaging
  • Aptamer labeling
  • DNA nanotechnology
  • Biosensor development

Every DY-549P1-modified oligonucleotide is synthesized using automated solid-phase phosphoramidite chemistry and is available with optional HPLC or PAGE purification. Comprehensive analytical quality control includes HPLC, LC-MS, MALDI-TOF, UV-Vis spectroscopy, and fluorescence verification to confirm successful dye incorporation and product purity.
