Cy5 dye is compatible with various instrumentation including many fluorescent microscopes, imagers, scanners, and fluorescence readers. This dye replace Alexa Fluor 647, and DyLight 649. It has an absorbance maximum of 646 nm and an emission maximum of 662 nm. This dye are suitable for use in application such as fluorescence resonance energy transfer (FRET), time resolved- fluorescence resonance energy transfer (TR-FRET), and fluorescence polarization (FP). Cy5 dye can be incorporated at any position within an Oligonucleotide post-synthetically or at time of synthesis. An oligonucleotide modified with a primary amine group is used to covalently attach dye with oligonucleotide via NHS ester chemistry; other cross-linking chemistries are also available depending on particular project specifications . Dual HPLC purification is highly recommended for post-synthetic conjugation.
