Bio-Synthesis offers
photocleavable PC amino C6 oligo modification at various
scales. PC Amino C6 containes a non-nucleosidic moiety that can be used to incorporate
a UV photo-cleavable primary amino group onto the 5’-end of an oligonucleotide.
The amino group is separated from the 5’-end nucleotide base by the photo-cleavable
group and a 6-carbon spacer arm to minimize steric interaction between the amino
            group and the oligo
1. The presence of the amino group allows the user
to end label the oligo with a variety of different affinity, reporter or protein,
as well as immobilization on an activated solid support, depending on the application.
The photocleavable group can be selectively cleaved by illumination with UV light
at 300-350 nm spectrum, thereby releasing the marker moiety or detachment of the
oligo from the solid support1. The released oligo contained a 5-phosphate.

PC Amino C6 has been used in peptide-DNA conjugates as photo-cleavable mass marker
(PCMM). The experiment uses a PC DNA oligonucleotide to function as the hybridization
probe and the peptide is then released by UV photo-cleavage during the ionization/desorption
phase of UV-MALDI, and thus serves as a mass tag that identifies the presence of
            the target sequence2.

PC Amino C6 is also used to create “caged” oligonucleotides where oligonucleotide
activity is suppressed until released by an external factor (such as UV light).
Caging oligonucleotides would provide new possibilities for controlling biological
            mechanisms3.

            Purification:

Photocleavable PC amino C6 modified oligonucleotide can be complished by various
methods. Desalting or cartridge purification is acceptable, however, additional
purificaiton by HPLC is strongly recommended.

        Cleavage Protocol

Optimal cleavage is obtained with exposure to long-wave UV light in the 300-350
nm spectral range. Cleavage releases the oligo with a 5'-phosphate group.
