Polyhistadine tag removal

For selective removal of the tag a cleavage site such as the sequence recognition motif for the TEV protease. Enzyme cleavage site needs to be inserted into the expression vector used for protein expression. If no selective sequence motif or site is present it is almost impossible to remove the tag. Hence, most recombinant proteins that are sold of the shelf actually contain the tag. The tag may interfere with some experiments if protein-protein interactions are planned to be studied.
