A main reason is that the synthesis columns may not be loaded with exactly the same amount of CPG. For example, a 200 nmole column has a minimum of 200 nmols of 3’ base and sometimes it may very well have more than this amount. The scale of synthesis is always a starting scale of synthesis and does not reflect the the final yield.
