Telomeric PNA FISH on Metaphase Over the last decade, nucleic acids have been extensively modified, by replacing the phosphodiester linker or the sugar-phosphodiester backbone by various neutral or charged structures. Several of these modified oligonucleotides have improved properties in terms of affinity and binding to DNA and RNA. Two significant representatives of this new type of oligomers are locked nucleic acids (LNAs), also known as bridged nucleic acids (BNAs), and peptide nucleic acids (PNAs). Peptide Nucleic Acid (PNA) like LNA, is a DNA analogue of much recent interest. Structurally these analogues are very different, however their application is in many respects very similar. Each technology has its own advantages and choice between them principally depends upon the experimental conditions and specifics of the application.
