The linear ssDNA precursor is treated with Exonuclease to remove all linearssDNA. Therefore, the amount of residual linear ssDNA is minimal if not zero. However, when ssDNA has a strong secondary structure, it is possible that Exonuclease treatment is incomplete. In this case, there is a possibility that some linear ssDNA is still in the sample.
