What is a readout probe?

A "readout probe" can be any oligonucleotide probe that allows for the detection or readout of a complementary target sequence.

Readout probes are useful tools for in situ hybridization (ISH), fluorescence in situ hybridization (FISH), single-molecule fluorescence in situ hybridization (smFISH), multiplexed error-robust fluorescence in situ hybridization (MERFISH), or sequencing-based approaches.

Flurecently labeled readout probes enable transcriptome analysis in single cells. Techniques called single-molecule fluorescence in situ hybridization (smFISH) and multiplexed error-robust fluorescence in situ hybridization (MERFISH) utilize these types of probes.

Fluorescently labeled readout probes contain sequences complementary to readout sequences in targeted RNA with a Cy5 dye attached at the 3’ end.
