To control the coupling efficiency, online trityl monitoring of all oligonucleotides were carefully checked each cycle. After synthesis, all oligonucleotides quantity are analyzed by optical density (OD260) measurement. Randomly selected unmodified and all modified oligonucleotides are identified by MALDI-TOF mass Spectrometry, and purity check by PAGE or HPLC.
