ATTO® 390 Fluorescent Dye Oligonucleotide Labeling

ATTO® 390 is a premium blue fluorescent dye developed for high-performance fluorescence imaging and analytical applications. The dye exhibits excellent brightness, outstanding photostability, and high fluorescence quantum yield, making it an excellent choice for fluorescence microscopy, molecular diagnostics, and bioanalytical research requiring stable blue fluorescence.

ATTO® 390 is commonly conjugated to DNA, RNA, PNA, LNA, siRNA, antisense oligonucleotides (ASOs), aptamers, peptides, proteins, and other amino-modified biomolecules using NHS ester chemistry, forming a stable amide bond with primary amino groups. Alternative site-specific conjugation methods, including click chemistry, can also be employed depending on the experimental design.

The dye exhibits excitation and emission maxima of approximately 390 nm and 479 nm, respectively, making it compatible with 375 nm, 385 nm, and 405 nm laser systems commonly found on fluorescence microscopes, confocal microscopes, imaging systems, and flow cytometers. Its blue emission provides excellent spectral separation from green, orange, red, and near-infrared fluorophores, making it well suited for multicolor fluorescence experiments.

ATTO® 390 can be incorporated at the 5′ end, 3′ end, or internal amino-modified positions, and may be combined with numerous additional modifications including quenchers, biotin, thiol modifiers, amino modifiers, PEG linkers, click chemistry handles, GalNAc, cholesterol, peptides, lipid conjugates, and other custom chemistries.

Each labeled oligonucleotide is synthesized under stringent quality standards and is available with optional HPLC purification, together with analytical characterization by HPLC, LC-MS, MALDI-TOF, UV-Vis spectroscopy, and fluorescence analysis to verify labeling efficiency, sequence identity, and product purity.

Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at 20°C) in a nuclease-free environment should be stable for years.
